چکیده

OBJECTIVES To characterize the T cell subsets producing interleukin 17 (IL-17) isoforms A and F in hypertensive kidneys and vessels and determine whether inhibition of IL-17 signaling lowers blood pressure and end-organ damage in a mouse model of hypertension. BACKGROUND T cell derived cytokines play a central role in the pathophysiology of hypertension and contribute to end-organ dysfunction. We previously showed that mice genetically deficient in IL-17A exhibited blunted hypertension and reduced renal and vascular dysfunction in response to angiotensin II (Ang II) infusion. Monoclonal antibodies to IL-17 isoforms or the IL-17 receptor are emerging as novel therapeutics for psoriasis and related autoimmune disorders. METHODS AND RESULTS Mice were infused with Ang II for 4 weeks to induce hypertension. Using flow cytometry and intracellular staining, we determined that the primary T cell subsets producing IL-17A and IL-17F in the kidney and aorta are gamma delta (γδ) T cells and CD4+ T helper 17 (TH17) cells. Monoclonal antibodies were administered twice weekly starting 2 weeks after the onset of Ang II infusion. Antibodies to IL-17A or the IL-17 receptor A subunit (IL-17RA), but not IL-17F, lowered blood pressure by 30 mmHg, attenuated renal and vascular lymphocyte infiltration, and reduced renal transforming growth factor beta (TGFβ) levels (a marker of renal fibrosis) compared to control IgG1 antibodies. Inhibition of IL-17 signaling also blunted the progression of albuminuria. CONCLUSIONS Monoclonal antibodies to IL-17A or IL-17RA, but not IL-17F, may be a useful adjunct treatment for hypertension and the associated end-organ dysfunction.

Deficiency of the interleukin 17/23 axis accelerates renal injury in mice with deoxycorticosterone acetate+angiotensin ii-induced hypertension.

T cells participate in angiotensin II (Ang II)-induced hypertension. However, the specific subsets of T cells that are important in the end-organ damage are unknown. T-helper 17 cells are a recently identified subset that produces interleukin 17 (IL-17) and requires interleukin 23 (IL-23) for expansion. To evaluate the role of the T-helper 17 immune response in hypertensive renal and cardiac en...

Interleukin-17A and Interleukin-17F mRNA Expressions in Peripheral Blood Mononuclear Cells of Patients with Multiple Sclerosis

Bakground: Multiple sclerosis (MS) is a CD4+ T cell-mediated autoimmune disease affecting the central nervous system (CNS). It was previously believed that Th1 cells were pathogenic T cells in experimental autoimmune encephalomyelitis (EAE). However, the functional role of Th1 cells in EAE has been reconsidered upon the discovery of IL-17- producing T cells which are consider as dominant effect...

Crocin prevents acute angiotensin II-induced hypertension in anesthetized rats

Objective: Angiotensin II (Ang II), the main product of renin-angiotensin system (RAS) has a well-known role in cardiovascular regulation. Over-production of Ang II is one of the important underlying mechanisms of hypertension. In this study, the effect of crocin on cardiovascular responses in rats with acute hypertension induced by Ang II was evaluated. Materials and methods: Rats were divided...

Inflammation and mechanical stretch promote aortic stiffening in hypertension through activation of p38 mitogen-activated protein kinase.

RATIONALE Aortic stiffening commonly occurs in hypertension and further elevates systolic pressure. Hypertension is also associated with vascular inflammation and increased mechanical stretch. The interplay between inflammation, mechanical stretch, and aortic stiffening in hypertension remains undefined. OBJECTIVE Our aim was to determine the role of inflammation and mechanical stretch in aor...

Introduction and purpose: One of the complications of overweight and obesity is inflammation and hypertension. This study aimed to evaluate the effect of eight weeks of aerobic training with quercetin supplementation on hypertension, CRP, and IL-6 in inactive overweight women. Materials and Methods: Forty inactive healthy overweight women (age 33.25±6.1 years, fat percentage 36.65±2.78 and bod...
